Developed oral medication, one pill a day… Target for FDA application in 2027 and approval in 2028. Commercialization profits reinvested in follow-up new drugs... Target of 1 Trillion Won in sales by 2030
It takes a huge amount of money and a long time to lead a new drug directly to global phase 3 clinical trials, but the experience gained in the process becomes an important asset for the company. This is because the capabilities secured through multi-country clinical operations, data management, and approval preparation can be used for follow-up new drug development. The Market Times takes a look at domestic bio companies and new drug candidates directly challenging global phase 3 clinical trials, and points out the process toward commercialization from clinical trials to preparation for approval and production.
The first company is AriBio, which is developing ‘AR1001’, an oral Alzheimer’s disease treatment candidate. A global phase 3 clinical trial involving 1,535 patients with early-stage Alzheimer's disease in 13 countries has been completed and the main results are scheduled to be released in November. The company has begun preparing New Drug Application (NDA) data and production, and aims to apply for approval from the U.S. Food and Drug Administration (FDA) in 2027 and approval in 2028.
November results released... Challenge for first-line treatment for early-stage Alzheimer's disease
AR1001 targets early Alzheimer's disease like Lekembi and Kissunla, but its mode of action is different. While the two antibody treatments directly target amyloid beta, AR1001 is being developed with the goal of inhibiting the activity of the PDE5 enzyme to induce various effects such as improving cerebral blood flow, protecting nerve cells, and inhibiting hyperphosphorylation of tau protein.
The key to commercialization is the results of the global phase 3 clinical trial ‘POLARIS-AD’. The main results will be revealed for the first time at ‘CTAD 2026’, an Alzheimer’s disease clinical trial conference held in Boston, USA on November 16, 2026. Fred Kim, head of AriBio's U.S. branch, explained that the timing of the announcement was decided after consulting with partners such as Fosun Pharmaceuticals in consideration of the academic society's policy of restricting prior disclosure.
The company announced that the dropout rate in phase 3 clinical trials was 12.2% and the participation rate in extension trials was 95.5%. Director Kim said that this figure shows how consistently patients participated in clinical trials, and emphasized that the experience and capabilities accumulated while conducting global phase 3 clinical trials will be assets for the development of follow-up new drugs.
In the outcome analysis, attention is paid to the treatment effect not only for the entire patient group but also for the single administration group. The goal of the first approval is to secure evidence to use AR1001 as a first-line treatment for early-stage Alzheimer's disease. Director Kim explained that in order to develop a treatment that can be used from the stage of mild cognitive impairment due to Alzheimer's disease, it is important to confirm meaningful results even in the single administration group.
From data verification to production… Parallel preparation for FDA approval
Efforts were also made to increase the reliability of data to be used in permit applications. The company utilized clinical trial institutional investors (CROs) in each field and conducted evaluator training and cross-verification of data. The explanation is that the U.S. branch had its own clinical trial monitoring personnel, established an internal management system, and directly participated in clinical trials and data inspection.
Director Kim also presented examples of cooperation with Fujirebio, a diagnostic device developer, as evidence for data quality. He explained that in the process of obtaining FDA approval for Fujirebio's Alzheimer's disease blood diagnostic device, the quality of the AR1001 phase 3 clinical data was also indirectly verified.
AriBio is preparing NDA materials with partners along with data verification and analysis. The production division announced that it has completed the manufacturing of the production batch required for the license application and has entered the stage of evaluating its stability. We plan to utilize contract development and manufacturing organizations (CDMOs) in India and the United States for commercial drug production, and gradually expand production in China after approval and sales commencement.
Target of 1 Trillion Won in sales by 2030... Reinvest in follow-on new drugs
The company expects that profits from the contract will increase as these preparations lead to commercialization. Branch Manager Kim revealed that the down payment and option consideration received through the copyright/option contract exceeds 1,500 Billion Won. The goal is to achieve 1 Trillion Won in sales by 2030 by securing step-by-step compensation and sales profits according to meeting contract conditions such as future licensing and sales. The plan is to reinvest this in the development of follow-up new drugs and lay the foundation for continuing research and development with its own sales.
Research and development will be conducted in parallel with expanding the indications for AR1001 and securing a follow-up pipeline. Starting with early-stage Alzheimer's disease, AR1001 plans to expand its scope of application to include stroke, Parkinson's disease, vascular dementia, and traumatic brain injury. Combination therapy combining with other treatments is also undergoing preclinical research, and details will be disclosed later in consideration of patents and business strategies.
Among the follow-up candidates, ‘AR1005’, a treatment for Lewy body dementia, is undergoing phase 2 clinical trials and aims to announce major results in 2027. The development of ‘AR1004’, a candidate for treatment of mild cognitive impairment, which has been approved for phase 2 clinical trial (IND), is also continuing. In addition, we plan to use our own artificial intelligence (AI) platform ‘ARIDD’ to discover new candidate substances, review the introduction of technology for drugs that act on multiple mechanisms, and expand the pipeline.
In the process of expanding the scope of development, the capabilities of group companies are also utilized. By adding AriBio's global clinical and licensing experience to AriBio Lab's immune and neuroinflammation research capabilities, we will pursue the development of an Alzheimer's vaccine and review candidate substances for combination therapy. We are collaborating with AriBio Holdings to build an AI data infrastructure that will utilize research and clinical data.
AriBio is developing AR1001 so that patients can continue treatment at home by taking one pill a day. Through this, the plan is to lower the burden of hospital visits for patients and their families and, in the long term, to enable use in primary healthcare Institutional Investors and areas with low access to medical care. Director Kim said that 16 years of research and development is about to come to fruition, and that he will carefully review the clinical data, disclose the results as is, and prepare for follow-up development.
